Mourinho handed a recall to Chris Smalling, while Henrikh Mkhitaryan, Nicolo Zaniolo and Bryan Cristante also returned to the Italian side's line-up.
The Dutch outfit took control in the early stages of the encounter, while Roma struggled to get any possession in the opening 15 minutes. Mourinho was forced to make change 17 minutes in when Mkhitaryan was forced off with an injury and replaced by Sergio Oliveira. Despite the sluggish start, Roma took the lead on 32 minutes when Zaniolo got on the end of Mancini's cross before poking the ball beyond the on-rushing Justin Bijlow.
Feyenoord nearly bagged an equaliser four minutes before the break when Orkun Kocku did well to pick out Guus Til with a ball over the top, but the striker was denied by Rui Patricio.
The Dutch side went even closer shortly after the break when Mancini's attempted clearance from Kocku's cross crashed against the upright before Patricio denied Til on the rebound.
With Feyenoord taking control, Mourinho responded with a double change after the hour mark as Jordan Veretout and Leonardo Spinazzola replaced Zaniolo and Nicola Zalewski.
Lorenzo Pellegrini nearly wrapped up the win with four minutes left on the clock after getting on the end of Veretout's pass, but the midfielder's strike was palmed away by Bijlow.
Feyenoord everything forward in search of an equaliser, but the Serie A side remained resolute at the back and held to clinch the first-ever Conference League title.
