Xavi Hernandez made four changes from the loss to Eintracht Frankfurt as Sergino Dest, Clement Lenglet, Frenkie de Jong and Memphis Depay were all recalled.
To makes matters worse, it was also confirmed that young midfielder Pedri will miss the rest of the season due to a hamstring injury.
The home side were eager to bounce back from Thursday's loss, but only managed to carve out one chance in the opening 20 minutes after Ferran Torres was denied by goalkeeper Jeremias Ledesma. The Camp Nou was nearly silenced on 27 minutes when Lucas Perez was played through on goal by Sobrino's pass, but the former Arsenal forward sliced his strike wide of Marc-Andre ter Stegen's upright.
Other than long-range effort from Dembele before the break, it was a relatively disappointing opening 45 minutes for the Catalan giants.
That all changed in the second half as Cadiz broke the deadlock three minutes after the break when Ter Stegen parried Sobrino's header into the path of Perez to fire into an empty net.
Xavi responded by throwing on Pierre-Emerick Aubameyang for Depay just after the hour mark, while Cadiz brought on Alejo and Fede.
The visitors had a chance to double their lead with 14 minutes left on the clock as Alejo sent a teasing cross into the box that found Alex, who steered his strike wide of the mark.
Barcelona threw everything forward in search of an equaliser, with Aubameyang going closest on 89 minutes with a volley that was brilliantly saved by Ledesma, as Cadiz held on for their first-ever win at the Camp Nou.
